Are there any programs running in the background using it? Try and find AMCAP on your PC. If you find it, open it. It will show your webcam image. If it works, you know that the webcam itself is working. Then, close it. Try Skype again, if it doesn't work, back up AMPCAP to disc or something, and delete it. It's only an EXE file and doesnt appear to have any system files.
If it still doesn't work you may or may not wish to put the file back. I had problems with cam until I deleted it. I reinstalled it from CNET and my webcam was fine after that for msn and skype.
